Raja Rani is about John, a young man, and Regina, a woman, who get married, even though they can't stand each other and how they realize that, there is life and love in their new relationship. Both have a romantic past - she with the innocent Jai and he with the chirpy Nazriya - have lost their loved ones in different ways, and are unwilling to come out of their loss. How they decide to let go of the past and embrace the future is what the plot is all about.

He takes over to fulfill his duties as a doting son and extends it by sacrificing his love. The rest of the story is about the transition of a young man to a leader.

The superstar Gautham Krishna (Prakash Raj) is set to release his new film, Traffic Constable Satyamoorthy (Cheran) is to join back on duty, the day is special for Dr. Robin (Prasanna) who is celebrating his first wedding anniversary, Karthik Vishwanathan (Sachin) an aspiring television journalist, is about to interview Gautham Krishna the very same day. All this comes to a standstill when Karthik travelling on a bike is hit by a speeding car and is rushed to the hospital. Karthik is in state of comatose and is being kept alive on ventilator. Meanwhile, Gautham's ailing daughter has a heart condition and is in deteriorating state, requiring urgent heart transplant. At first Karthik's parents are hesitant to let him off the ventilator, with much persuasion by Karthik's girlfriend (Parvathi Menon) they agree. The heart organ is available now, but there is a problem of transporting it from Chennai to Vellore in a short period of time. The people volunteering for this transportation of organ, are to take up this impossible task. After having killed the notorious drug dealer Mayil Vaaganam, Durai Singham (Suriya) has gone undercover and is working as an NCC officer in a school in Thoothukudi. Raghu (Jai) and his parents move from Royapuram to Visalakshi Thottam, having pincode 600028. He is a member of the Royapuram Rockers Cricket team and not very excited at the prospect of moving and residing in the same area as his rivals (Sharks). He is replaced by the new guy in the team for the recent matches. A love triangle unfolds between Karthik (Siva), Selvi (Vijayalakshmi) and Raghu. Karthik confronts this with Raghu, but he tells him that she in fact loves him. He slowly even befriends the other Shark team members and joins their team, determined to beat the Rockers in the upcoming tournament. Pazhani's sister is the cause of argument between them. Eventually, the game and friendship is placed above the relationships. In an unfortunate event, Karthik is stabbed by his fixer brother's (Sampath) enemies and is hospitalized.
